What is PT-141?
PT-141 is the development code for bremelanotide, a synthetic analogue of alpha-melanocyte-stimulating hormone (alpha-MSH). It is a cyclic heptapeptide: Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH, with a lactam bridge between the aspartate and lysine side chains, an acetylated N-terminus, norleucine in place of methionine and one D-amino acid (D-phenylalanine).
It is a close relative of Melanotan II. The two share the same ring and sequence and differ only at the C-terminus: PT-141 ends in a free carboxylic acid, Melanotan II in an amide. Researchers at the University of Arizona designed Melanotan II; Palatin Technologies later developed PT-141 from it.

Published research on PT-141
PT-141 has a longer clinical record than most compounds sold for research, because it was developed as a medicine. The early human studies gave it intranasally; the later phase 2b and phase 3 programmes, and the approved product, used subcutaneous injection.
- Pharmacology overview (2003). Palatin’s researchers described PT-141 as an agonist at the MC3 and MC4 receptors and summarised its effects in rats, nonhuman primates and early human studies. PT-141: a melanocortin agonist for the treatment of sexual dysfunction. Molinoff PB, et al. Annals of the New York Academy of Sciences, 2003.
- Intranasal phase 1 (2004). Two double-blind, placebo-controlled studies of intranasal PT-141 reported dose-dependent exposure, a median time to peak of 0.5 hours and a mean half-life of about 1.9 to 2.1 hours. Flushing and nausea were the most common adverse events. Double-blind, placebo-controlled evaluation of the safety, pharmacokinetic properties and pharmacodynamic effects of intranasal PT-141, a melanocortin receptor agonist, in healthy males and patients with mild-to-moderate erectile dysfunction. Diamond LE, et al. International Journal of Impotence Research, 2004.
- Central mechanism in rats (2004). In female rats, PT-141 selectively increased solicitational behaviour without changing other measured behaviours or general motor activity, consistent with an effect on central melanocortin receptors. Selective facilitation of sexual solicitation in the female rat by a melanocortin receptor agonist. Pfaus JG, et al. PNAS, 2004.
- Phase 2b dose-finding (2016). A 12-week randomised, placebo-controlled trial in 327 premenopausal women (NCT01382719) reported significant differences from placebo on its primary endpoint for the pooled higher-dose groups. Bremelanotide for female sexual dysfunctions in premenopausal women: a randomized, placebo-controlled dose-finding trial. Clayton AH, et al. Women’s Health, 2016.
- Phase 3, RECONNECT (2019). Two identical 24-week randomised, double-blind trials (NCT02333071 and NCT02338960, 1,267 women randomised) reported significant differences from placebo on both co-primary endpoints. Nausea, flushing and headache occurred more often with bremelanotide. Bremelanotide for the Treatment of Hypoactive Sexual Desire Disorder: Two Randomized Phase 3 Trials. Kingsberg SA, et al. Obstetrics and Gynecology, 2019.
- 52-week extension (2019). The open-label extension of RECONNECT reported no new safety signals; nausea (40.4%), flushing (20.6%) and headache (12.0%) were the most common treatment-related events. Long-Term Safety and Efficacy of Bremelanotide for Hypoactive Sexual Desire Disorder. Simon JA, et al. Obstetrics and Gynecology, 2019.
Regulatory status
Bremelanotide is FDA-approved as Vyleesi (bremelanotide injection, NDA 210557, approved 21 June 2019), a subcutaneous autoinjector for a specific indication in premenopausal women. That approval covers Vyleesi only. PT-141 sold as a research material, including this spray, is not an approved drug and is for laboratory research only. Bremelanotide: First Approval. Dhillon S, et al. Drugs, 2019.

QuestionsPT-141 Spray: frequently asked
What is PT-141?+
PT-141 is bremelanotide, a synthetic cyclic heptapeptide analogue of alpha-MSH that acts at melanocortin receptors. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH.
Is PT-141 the same as bremelanotide?+
Yes. PT-141 is the development code; bremelanotide is the nonproprietary name. The FDA-approved product is called Vyleesi.
What is in the PT-141 nasal spray bottle?+
20 mg of PT-141 in 20 mL of solution. The current lot’s certificate confirms identity by HPLC-DAD and reports the measured concentration.
How is PT-141 different from Melanotan II?+
They share the same cyclic sequence. PT-141 ends in a free carboxylic acid (C50H68N14O10) and Melanotan II in an amide (C50H69N15O9).
Was PT-141 studied as a nasal spray?+
Yes. The early human studies, including a 2004 phase 1 programme, gave PT-141 intranasally. Later phase 2b and phase 3 trials and the approved product used subcutaneous injection.
What is the CAS number for PT-141?+
CAS 189691-06-3 (PubChem CID 9941379).
